To analyze the impact of Doxycycline on the efficacy and safety of doxycycline-containing formulations of veterinary pharmaceuticals. Methodology
We conducted a retrospective, prospective, multicenter study using a global population based survey using the same survey instrument in the United Kingdom. Study sample
The study population consisted of 4200 veterinary patients aged 20 to 65 years who were admitted with a diagnosis of chronic obstructive pulmonary disease (COPD), with or without previous treatment with doxycycline for 5 to 10 years. In the following 7 years, the patients were randomly assigned to either a doxycycline-containing formulation (DoxyC, 1 mg/kg body weight in 0.5 mL) or a control group (DoxyP, 0.5 mg/kg body weight in 0.5 mL). The primary endpoint was the change in COPD symptom score from baseline to the end of the study. In the first 7 days, the patients were assessed with a spirometry test and the severity of COPD symptoms, including dyspnea and dyspnoea, and a pulmonary function questionnaire, as well as a patient-reported global assessment of the disease activity (PROXIMAX).
Results
The number of patients was 1,083, with a median follow-up of 5.2 months. There were 1631 (49.8%) DoxyP-treated and 1,006 (28.8%) DoxyC-treated patients in the study and 1292 (73.1%) patients in the control group. There were significant differences between the groups in mean change in the severity of symptoms, dyspnea, and dyspnoea from baseline to end of the study (p < 0.05). The patients who were DoxyP-treated had significantly lower mean severity of symptoms (p < 0.05) and dyspnoea than those in the control group (p < 0.05). DoxyP-treated patients had significantly lower mean score in dyspnoea (p < 0.05) and more severe dyspnoea (p < 0.05) than those in the control group.
Conclusion
Doxycycline was found to be a very effective treatment for COPD when used in a dose-dependent manner in a dose-time dependent manner. The results suggest that a dosage of 5 mg/kg body weight in 0.5 mL oral formulation may be able to reduce dyspnoea and improve COPD symptom severity and quality of life.
